Defective T cell proliferation upon stimulation of plate-bound anti-CD3. (A) Splenocytes from Rac2−/− and control mice were stimulated with varying amounts of plate-bound anti-CD3. WT, wild-type. (B) Purified CD4 T cells stimulated with a dose range of plate-bound anti-CD3. (C) Purified T cells were stimulated with plate-bound anti-CD3 (5 μg/ml) alone, in the presence of IL-2 (50 μ/ml), or plate-bound anti-CD28 (2.5 μg/ml). (D) Purified B cells were stimulated with anti-IgM (10μg/ml), anti-IgM (10 μg/ml) + IL-4 (1,000 U/ml), or LPS (1 μg/ml). [3H]Thymidine was pulsed 48 h after stimulation and harvested 12 h later.
