Ag-specific responses by transfectants expressing RT1-TCR-α and various TCR-β chains expressing random CDR3β. Vβ8+ TCR-β chains were randomly isolated from naive CD8+ cells from wild-type BALB/c mice. Their Vβ usage was Vβ8.1, the Jβ usage was Jβ2.1, 2.2, 2.4, and CDR3β sequences were random. These TCR-β genes were transfected into RT1-TCR–expressing TG40 cells by retrovirus-mediated gene transfer, and these transfectants were stimulated with irradiated splenocytes and P18IIIB (right) or immobilized anti–TCR-β Ab, H57 (left) for 24 h, as for Fig. 1. The concentration of IL-2 was measured by ELISA and the data are presented as the percentages of that of RT1-TCR-β-transfectant (RT1α).
