In vivo uptake of PPV-VLPs by DC subpopulations. C57BL/6 mice were intravenously injected with 50 μg Alexa 488–PPV-VLPs-OVA. 90 min or 15 h later their CD11c+ spleen cells were purified and stained with PE and APC-coupled mAbs for analysis on a FACScalibur® cytometer. Noninjected mice were used as control. (A) CD11c+ spleen cells from control mice were stained with APC–anti-CD8α and either PE–anti-CD11c (left) or PE–anti-CD4 (right). In this representative experiment, CD11c+ purified spleen cells contained 53% CD4+ CD8α−, 17% CD4− CD8α+, and 20% CD4− CD8α−. Similar values were obtained from injected mice.
