Analysis of the LACK-specific CD4 T cell response to lack-targeted L. major. 2 × 106 wild-type or lack-targeted parasites were inoculated into the hind footpads of BALB/c mice. After 96 h, lymphocytes from the draining popliteal lymph nodes were assayed for expansion of LACK-specific CD4 T cells using MHC class II I-Ad/LACK peptide tetramers as previously described (reference 6).
