Membrane TNF Expressing B2.20 Cells Induce Apoptosis but Not HIV Replication in ACH-2 Cells

ACH-2 cells were seeded at 125 × 103 cells/well, in the absence or presence of sTNF (30 ng/ml) or in wells containing monolayers of CHO or B2.20 cells as described in Materials and Methods. 22 h after onset of cultures, 10 μl of supernatant was removed and RT activity determined. Thereafter, the nonadherent ACH-2 cells were removed from the wells (60 wells for each experimental condition), pooled, and counted.

The cells were further stained with YOPRO-1™, 200 cells for each group were counted and cells displaying fragmented or condensed nuclei were scored as apoptotic.
