Results from a representative experiment are shown. Splenic T cells were purified and 10⁶ cells were stimulated in culture for 18 h with PMA (10 ng/ml) and ionomycin (1 μM) (PMA+I) or plate-bound and anti–CD3-ε. Supernatants were serially diluted and tested by ELISA as described in Materials and Methods. Concentrations were determined by comparison with known concentrations of cytokines provided by the manufacturers of the monoclonal antibodies used for ELISA.
