CD8⁺ T cells recovered from CD3fos-F(ab′)₂– treated F5/RAG-1⁻β2m⁻ thymuses are functionally mature CTL precursors. CTL activity shown is of cells recovered from CD3fos-F(ab′)₂–treated F5/ RAG-1⁻β2m⁻ thymuses (circles) or medium control thymuses (triangles). Thymocytes recovered from equivalent numbers of cultured lobes/group were stimulated with NP68 peptide-pulsed DCs for 4 d and tested for cytolytic activity on peptide-loaded (filled symbols) or control (open symbols) EL-4 target cells in a standard 4-h ⁵¹Cr–release assay. The effector ratio is presented as responder dilution as cells recovered from stimulation cultures were resuspended to the same volume.
