KREPA6 repression leads to loss of 20S editosomes. RNAi-A6 cell lines were grown in the (A) absence or presence of tet for (B) 2 d or (C) 4 d. Equal numbers of cells were lysed and fractionated on glycerol gradients. Odd-numbered fractions were analyzed by Western blot with a mixture of monoclonal antibodies specific for editosome proteins KREPA1, KREPA2, KREL1, and KREPA3, and a polyclonal antibody to KREPA6. Blots were also probed with a monoclonal antibody recognizing a component (2-OGDCp) of the mitochondrial 2-oxoglutarate dehydrogenase complex as a control.
