A volumetric reconstruction based on an 50-image stack of an excised mouse olfactory bulb (same sample as in Fig. 3) acquired with standard TPEF microscopy (a) and with 2π spiral phase DA-TPEF microscopy (b). Raw images were acquired every 2 μm to a depth of 150 μm. Lateral field of view is ∼1.3 × 1.3 mm² (Olympus 20 × NA = 0.95 water immersion objective). The laser wavelength was 800 nm and the power delivered to the sample was ∼20 mW. Volumetric reconstruction was performed with Amira software (ZIB, Berlin, Germany).
