Homophilic adhesion of MCF-7 cells to E-cadherin-Fc mutants E31A, KEG30-32RDT, and E31D. Serial dilutations of wild-type and mutated E-cadherin and IgG1 were immobilized on plates coated with goat anti–human IgG antiserum. Homophilic adhesion assays with E-cadherin–expressing MCF-7 cells were performed in HBS with 1 mM CaCl2, 50 mM glucose, and 0.1% BSA, as described in Materials and Methods. Results are shown as the mean percentage of MCF-7 cells bound ± 1 SD and are representative of at least two independent experiments in which each condition was performed in triplicate.
