Effect of duration of preincubation with DIDS on valinomycin- induced net efflux at 23°C (A) and at 37°C (B). Washed cells at 1.2 percent hematocrit in medium containing 1 mM KCl and 149 mM NaCl were pretreated with 10 μM DIDS for either 1 min (circles) or 30 min (squares), followed by addition of 3 μM valinomycin (at arrows), and the mean cell volume (MCV) was determined (see materials and methods) in duplicate suspensions (filled and hollow symbols) at the indicated times.
