Nef expression increases the number of IL-2-generating cells. Jurkat cells transduced with NL4–3 (triangles) and SF2 nef (diamonds), and neo (squares) and G2A (circles) controls were stimulated with anti-CD3 plus anti-CD28. Cell activation was examined by ELISPOT assay (A) and by in situ ELISA (not shown). Data from two input cell densities common to both the ELISPOT and in situ ELISA [5 × 10³ cells (open symbols) and 10⁴ cells (solid symbols)] are shown (B). The relationship between the number of activated cells and the total amount of IL-2 secreted for the various transductants at these cell densities was linear, r² = 0.98. Cells were transduced as in Fig. 1. Data represent mean determinations and SDs from triplicate wells.
