Activation of channel activity at +80 mV. Proteoliposomes reconstituted with PulD-PulS complex were allowed to fuse with a planar lipid bilayer by an osmotic gradient. When channel activity was observed, the membrane potential was clamped at +80 mV, and the channel activity was monitored for 10 min. Solutions in the cis and trans compartments were 10 mM Hepes-KOH, 400 mM KCl, pH 7.4, and 10 mM Hepes-KOH, 100 mM KCl, pH 7.4, respectively.
