FIG. 1.

Sequence analysis of CrRelish and schematic representation of expression constructs. (A) Schematic representation of full-length CrRelish and sequence alignment with the characteristic motifs of other NF-κB proteins. Full-length CrRelish contains an N-terminal RHD (gray box) with a conserved DNA-binding motif, followed by an NLS (black box) and a C-terminal IκB-like domain which contains six ANK repeats (white boxes). The amino acid sequences of NLSs from different organisms were aligned, and the first and last amino acids of the DNA-binding motif and the NLS are indicated. (B) Amino acid identity of RHD and ANK repeats between representative members of the p100 protein family. For a list of GenBank accession numbers of the sequences used, see Table S2 in the supplemental material. (C) Schematic representation of the expression vectors used in transfection experiments: full-length CrRelish, RHD+NLS+L (RHD with the NLS and the linker between the NLS and ANK), RHD+NLS (RHD with the NLS), and RHD (RHD only).
