Analysis of Pl^A2β3 synthesis after transduction of human CD34+ cells. Immunoblot showing Pl^A2β3 synthesis in megakaryocytes derived from transduced CD34+ cells. Seventeen days after transduction and differentiation, 850 μg of each cellular lysate was immunoprecipitated with a αIIbβ3 complex-specific mAb, AP2, and detected with human anti-Pl^A2β3 alloimmune serum (Upper). The blot reprobed with a polyclonal anti-β3 antibody demonstrates the presence of Pl^A1β3 in the untransduced Control sample (Lower). Molecular mass markers are shown in kDa (left).
