Purified B cells (1 × 105) were stimulated with anti-CD40 (10 μg/ml) plus IL-4 (10 ng/ml). After 6 days, IgM and IgG1 concentrations in the culture supernatants were measured by ELISA. IgM and IgG1 from unstimulated cultures were undetectable. Three independent experiments are shown, and the differences between traf5+/+ and traf5−/− are statistically significant (P < 0.001.
