Circles symbolize amino acid residues (gray, human NKCC1; black, sNKCC1). AI-V-I-L(4.5) is a tm-4 sNKCC1 mutant with the substitutions S386, V387, I389, V391, and F393 → A414, 415, V417, I419, and L421. In the mutant GD-VL (5.3), V419, N420, T422, and V423 in tm 5 of sNKCC1 are replaced by the corresponding G447, D448, V450, and L451 of hNKCC1. In the shark L-V-I-A (7.7) mutant, the substitutions are I504, A507, A509, and V511 → L532, V535, I537, and A539 in the tm 7 of the sNKCC1 protein.
