(A) Immunofluorescence labeling of monolayers grown in the presence or absence of 850 nM insulin for 2 d. Monolayers were fixed, permeabilized, and labeled with antibodies against α-1, α-2, and α-3 isoforms of rat Na⁺-K⁺ ATPase (scale bar, 25 μm). (B) Proteins from monolayers treated with 850 nM insulin for 2 d were also extracted and analyzed by Western blot analysis using the same antibodies as indicated for immunofluorescence.
