Activity of Css IV on mutant sodium channels with neutralized positive charges in segment IIS4. Conductance-voltage relationships measured as described in materials and methods in control (closed circles) or in the presence of 200 nM Css IV without (open circles) or with (open squares) a +50 mV, 1–ms prepulse that preceded the test pulse by 61 ms for wild-type Na_v1.2a (A), R850Q (B), R853Q (C), R856Q (D), K859Q (E), and K862Q (F) sodium channels. The holding potential was −100 mV. The insets show sodium current traces evoked by a test pulse to −65 mV before and after a prepulse to +50 mV in the presence of Css IV.
