In this and subsequent figures, the patch was excised at the arrow, and the bars indicate the application of PIP2 (5 μg/ml, unless indicated) or different [ATP] (as shown). Solutions were switched by moving the patch from one lane of the oil-gate chamber to another. The lower dashed line indicates zero current. Iinitial and IPIP2, used to determine Po,zero (see materials and methods), are indicated by additional dashed lines. The inserts in this and Fig. 3 show expanded records from the first 50–100 s after patch excision, from which ATP sensitivity and open probability (NA method) were estimated.
