Atg1 kinase activity is important for Atg8 dynamics during starvation. Cells from the atg1Δ atg11Δ strain expressing GFP-Atg8 (HCY116) were transformed with plasmids encoding Atg1 and Atg1K54A,M102A (Atg1KM). The cells were grown in SMD medium and shifted to SD-N for 2 h before fluorescence microscopy. For time-lapse experiments, the cells were immobilized on SD-N medium containing 2% agar on concavity slides as described in Materials and Methods, and pictures were taken every minute. Images are shown over the 7-min time course.
