A Kv1.4 C-type inactivation–deficient channel mutant. C-type inactivation was removed from Kv1.4 with the K533V mutation. HEK 293 cells expressing Kv1.4 K533V ΔN147 were depolarized for 4 s at +60 mV, from a holding potential of −100 mV, in the ionic conditions described in the Figure. In 135K+ i/5K+ o, the K533V mutation attenuates the decay of outward current. In 135 Na+ i/o, the K533V mutation essentially abolishes any decay of outward current. Representative currents exhibiting C-type inactivation in WT Kv1.4ΔN147 are also shown for reference.
