An irreparable MeG-T mismatch provokes excision in HeLa nuclear extract. (A) Mismatch-provoked excision in O6BG-treated HeLa nuclear extract was scored by conversion of 5’ G-T or 5’ MeG-T heteroduplexes to a form resistant to SalI (cleavage site 15 bp from mismatch) as described in Materials and Methods. The MeG-T heteroduplex was pretreated with AGT as indicated. Results are the average of at least 3 determinations ± one standard deviation. (B) 5’ G-T, MeG-T or T-MeG heteroduplexes, which were pretreated or untreated with AGT as indicated, were subjected to mismatch repair in HeLa extract (Materials and Methods). Repair was scored by conversion to XhoI (G-T and MeG-T) or HindIII (T-MeG) sensitivity. Results represent the average of at least 3 experiments ± one standard deviation.
