APRT^- colonies that arose by HR were analyzed by Southern blotting (Figure 2C) to determine whether the 3′ HpaII site (Figure 2B) was methylated (Met) or unmethylated (Unmet).

One clone was found to be methylated at both HpaII sites shown in Figure 2B.
