Lymphocytes were obtained from spleens of C57BL/6J mice and stained with mAbs directed to CD8 and either (A, C) Vβ8.3 or (B, D) Vβ7. Vβ8.3+CD8+ or Vβ7+CD8+ T cell populations were single-cell sorted into 96-well plates, followed by RT reaction, nested PCRs and sequencing of positive PCR products. The CDR3β sequences were deduced from the known genomic sequence (Arden et al., 1995; Iwamoto et al., 1987; Saito et al., 1984) and the Jβ-regions were assigned according to Gascoigne et al. and Malissen et al (Gascoigne et al., 1984; Malissen et al., 1984).
