HepG2 cells were transfected with 4.0 μg of expression vectors harboring cDNA for specific MBDs or empty vector, and the whole cell extract (200 μg of protein) prepared 48 h post-transfection was subjected to Western blot analysis with specific antibodies. B and C, effect of overexpression of MBDs on rRNA promoter activity. HepG2 cells were cotransfected with pHrD-IRES-Luc with (4 μg) of the empty vector or pcMBD1, pcMBD2, pcMBD3, or pcMeCP2. After 48 h of transfection, firefly luciferase activity was measured in the cell extracts. Results are represented as pHrD-IRES-Luc (RLU2)/pRL-TK (RLU1) activity.
