Proliferation rate and cell cycle distribution. (A) OrfA expression does not significantly change cell proliferation rate. Cells were seeded at 105/ml and counted after 24, 48 and 72 hours using a hemocytometer. (B) 104-C1-GFP and 104-C1-GFP-OrfA cells exhibit similar cell cycle distribution. Cellular DNA was stained with propidium iodide, cell fluorescence was measured by flow cytometry and discrimination of cells in G0/G1, S and G2/M phases was performed using FlowJo software. 104-C1 arrested cells were exposed to aphidicolin and Hoechst 33342 in two subsequent overnight incubations.
