ACS5 mRNA accumulation. Intact 3-day-old etiolated
seedlings were incubated in 5 μM benzyladenine for the times
indicated; total RNA was isolated and analyzed by Northern blotting.
The blot was hybridized with an ACS5 probe, stripped,
and rehybridized with a β-tubulin probe as a loading control.
Hybridization intensities were quantified with a PhosphorImager
(Molecular Dynamics). The level of ACS5 expression was
normalized to that of β-tubulin and plotted. (Inset)

prepared from 3-day-old etiolated seedlings grown as follows: lane 1,
acs5-2 plants grown on MS medium; lane 2,

plus 5 μM benzyladenine; lane 4, wild-type (Col) grown on MS medium;
and lane 5, eto2 grown on MS medium. Note that lanes 4
