Levels of mRNA expressed in cells transfected with either a test construct β-globin (A) or FV (B) upon coexpression of hUpf1 by RT-PCR. Either β-globin plasmid or foamy proviruses were transfected, along with either wt or a DN mutant hUpf1 at a molar ratio of 1:1. Ter-39 is a PTC-containing β-globin construct (34). Levels of Gag and Pol proteins produced in wt FV or the GR (−)st mutant upon coexpression of hUpf1 were examined by Odyssey Western blot analysis. At 48 h posttransfection, cell lysates were prepared to run on 10% SDS-polyacrylamide gel electrophoresis. The transferred membranes were probed with anti-Gag (C) or anti-Pol (D) antibody for detection. GAPDH was used to normalize the samples.
