Scheme for genetic analysis of histone H3 and H4 mutants. Histone H4 mutations in combination with the histone H3 N-terminal deletion Δ4–30 were analyzed by shifting cultures from galactose- to glucose-containing media. By shutting off wild-type H3 and H4 synthesis under control of the GAL1–10 promoter (in plasmid A) it is possible to examine the effects of mutated H3 and H4 (produced from plasmid B) on cell growth and nucleosome assembly in glucose medium. ∗ indicates the various H4 mutations.
