The temporal profiles of PARP, Wee1 and CDC27 cleavages during apoptosis in Jurkat cells, with tubulin serving as a negative control. The percentage of cell death was measured by MTT assay (20); 2 × 106 Jurkat cells were treated with either 10 ng/ml anti-Fas antibody or 0.1 μM staurosporine for various time periods as indicated. Aliquots of the total cell lysates were subjected to SDS/PAGE, and immunoblotting was performed by using anti-PARP, anti-CDC27, anti-Wee1, and anti-tubulin antibodies.
