hCUL1 complements the cdc53ts mutant phenotype. A cdc53–2ts mutant strain was transformed with pTS161-CDC53 and pTS161-hCUL1 plasmids that allow controlled expression of CDC53 and hCUL1 from the galactose-inducible GAL1 promoter. The empty vector alone was used as a negative control. Serial dilutions 1/10 of the individual transformants were spotted on synthetic galactose medium and were incubated for 5 days at restrictive (33°C) and permissive (24°C) temperatures.
