MI-219 blocks intracellular MDM2–p53 interaction, activates the p53 pathway, and inhibits cell growth. (A) Cells were treated for 24 h with different compounds. Whole-cell lysates (WCL) were analyzed by Western blotting. (B) SJSA-1 cells were treated for 15 h with MDM2 inhibitors or MG132. WCL were analyzed by immunoblotting, and coimmunoprecipitation was performed. (C) Cells were treated for 15 h with MI-219, and a Taqman real-time PCR assay was performed. (D) Cell growth inhibitory activity was evaluated in a WST cell growth assay. Data are shown as mean IC50 values. Error bars represent SEM.
