The ciliary epithelial cells were incubated in culture medium containing 0, 0.1, 1, 10, 50 or 200 μg ml−1 ClC-3 antisense oligonucleotide with 20 μg ml−1 lipofectin for 48 h (termed antisense 0.1, 1, 10, 50 and 200, respectively). A, the whole-cell current traces of the volume-activated currents under different treatments in the NPCE cells cycled through the 200 ms voltage steps (±40, 0, ±80 mV; 4 s interval). B, the mean values of the currents in a group of NPCE cells, measured 10 ms after the start of the +80 mV step, under different conditions. The number of cells used in each condition was: control, n = 26 and for 0.1, 1, 10, 50 and 200 μg ml−1 antisense n = 6, 8, 5, 10 and 17, respectively. The currents were inhibited by the ClC-3 antisense oligonucleotide in a dose-dependent manner.
