Inhibition of peak and sustained I_K by SKF10047 in outside-out patches

A, I_K reduction in outside-out patches with the control pipette solution containing 2 mM ATP. B, I_K reduction in outside-out patches with 50 nM okadaic acid and 2 mM AMPPcP in the patch pipette (no ATP). Voltage pulses 300 ms in duration from -80 mV to 50 mV activated I_K. Averages of ten traces are shown (collected at 15 s intervals) for both predrug control and after 1 min exposure to SKF10047 (100 μm). I_K recovered to predrug levels within 1 min of drug removal (data not shown). I_K recorded from outside-out patches is nearly identical in time course but smaller in amplitude compared with typical whole-terminal I_K (e.g. Fig. 1).
