Cleavage of selected phage clones by KLK6. Purified phages were incubated with 0 or 0.1 μM KLK6 in 50 mM Tris-HCl, 150 mM NaCl containing 1 mM EDTA (pH 7.5) at 37°C for 45 min. The reaction mixture was separated with 4%–20% gradient SDS-PAGE and then analyzed with anti-Flag antibody by Western blotting. The label “+” or “−” indicates with or without enzyme, respectively. The octapeptide sequences are shown above the respective band. AAFRFSQA has been reported to exhibit the maximum rate of cleavage by KLK6 in vitro (Angelo et al. 2006).
