A, GITR stimulation decreased phosphorylation of Stat5 in
IL-15-treated NK cells. Purified NK cells were cultured in the
presence/absence of 10 ng/ml IL-15 in plates coated with the control IgG or
anti-GITR antibody for 10, 20, and 30 min. Cells were collected and processed
for Western blot analysis using anti-p-Stat5 and anti-p-p38 antibodies.
B, GITR stimulation decreased phosphorylation of Akt and PDK1 in
IL-15-treated NK cells. Purified NK cells were cultured in the
presence/absence of 10 ng/ml IL-15 in plates coated with control IgG or
anti-GITR antibody for 1, 2, or 3 days. Cells were collected and processed for
Western blot analysis using anti-p-Akt, anti-Akt, or anti-p-PDK1
antibodies.
