Effects of Ac-SDKP on ET-1-induced MAPK ERK1/2 phosphorylation in rat CFs. CFs were stimulated with ET-1 (1 nM) in the absence or presence of Ac-SDKP (10 nM) at 37°C for 5 min. Top: Western blots of phosphorylated and total ERK1/2. Bottom: semi-quantitative data as % of control from triplicate blots for each protocol. Note that ET-1 markedly increased ERK1/2 phosphorylation, which was attenuated by pretreatment of cells with Ac-SDKP (10 nM). Total ERK1/2 was not changed by any treatment and also served to confirm equal protein loading.
