(A) Localization of Rga4 during the cell cycle. rga4:GFP cells grown on EMM agarose were observed. Representative projection images are aligned in the order of cell cycle progression. (B) The Δpom1, pom1−2, and Δtea1 mutants show a sock-like localization pattern of Rga4. Rga4GFP was observed in Δpom1, pom1−2, Δtea1, Δbud6, Δtea3, and orb2−34 mutants. Midsection images are shown. Asterisks mark the cell ends from which Rga4GFP was excluded. Arrows indicate dividing cells. (C) Localization of Rga4 is altered by ectopic overexpression of Pom1 kinase. A rga4:RFP CRIB-GFP strain carrying the pREP1-pom1:myc plasmid was grown at 30°C in EMM medium without thiamine to overexpress Pom1 and observed at 14 hr and 16 hr after removal of thiamine.
