Reconstitution of uracil-initiated BER with the purified catalytic subunit of human pol γ. BER reactions (see Materials and Methods) were incubated for 10 min at 15°C, 25°C, or 37°C (lanes 1–3, respectively), and products were analyzed by denaturing PAGE and autoradiography. Incorporation of a single [α-³²P]dCMP residue produced a radiolabeled 22-mer intermediate (arrow), and combined dRP lyase and DNA ligase activities generated the 51-mer product (arrow).
