DCs migrate to the spleen. DCs were isolated from the bone marrow of DBA/1j mice and cultured in the presence of GM-CSF for 4 days. They were then transfected with AdGFP at 50 pfu/cell for 18 hours in vitro before being intraperitoneally injected into CII-primed DBA/1j mice. Forty-eight hours later, the spleen was collected and embedded with OCT. Ten-micrometer frozen sections were counterstained with Hoechst and examined by fluorescence microscope, then photographed.
