Neuro2a and HeLa cell lines transfected with our Tet-On-inducible system for the gene RAGE. To test the cell-type specificity of the neuronal-specific enolase (NSE) promoter, the cell lines were either co-transfected with set1 (pCMV-rtTA and pTREtight-RAGE-EGFP) or with set 2 (pNSE-rtTA and pTREtight-RAGE-EGFP) plasmids. After 24 h of transfection, RAGE expression was induced by supplementing the culture medium with 2 μg/mL of dox. EGFP expression was recorded using a nikon inverted microscope. 1 = no induction; 2 = cells co-transfected with set 1 plasmids and induced with 2 μg/mL of dox; 3 = cells co-transfected with set 2 plasmids and induced with 2 μg/mL of dox.
