Spleen cells isolated from the four groups boosted by TCI were restimulated in the presence of 0.1 to 10 μg/ml CRM197. Spleen cells isolated from the group boosted parenterally with DTxd vaccine were restimulated in the presence of 0.1 to 10 μg/ml of purified DTxd. ANOVA was performed for all three cytokines at each dose of restimulating antigen. At the highest dose of restimulating antigen, TCI boost with CRM197 with CT as the adjuvant significantly increased splenocyte production of IFN-γ compared to cells isolated from mice in the PBS control group (P < 0.05). Data are the means ± SEMs from three separate spleen cell cultures.
