WT TAg, but not the K1 mutant, induces phosphorylation of Akt in U2OS cells. (A) U2OS cells were pretreated with either 50 μM of LY (+) or the same volume of dimethyl sulfoxide (the LY solvent; −) for 1 h at 37°C and then mock infected or infected with recombinant adenoviruses (MOI of 5) in the presence or absence of 50 μM of LY. At 24 h postinfection, whole-cell lysates were prepared and evaluated by Western analysis using antibodies described in Materials and Methods. (B) The activation of Akt by TAg is not due to alterations in key protein levels.
