Transport of the bivalent QacA substrate DAPI mediated by QacA mutants in which tryptophan has been replaced with alanine or phenylalanine. E. coli DH5α cells expressing each mutant (identified above each plot) were loaded with 8 μM DAPI and monitored fluorimetrically after energization of transport. Transport reactions were conducted at least in duplicate, and the results of representative experiments are shown.
