Effects of intracellular calcium chelation by BAPTA on C2-ceramide- vs. bradykinin-activated eNOS translocation. BAEC that had been incubated with or without BAPTA/AM (20 μM for 20 min) were then treated with bradykinin (BK; 1 μM for 5 min) or C2-ceramide (5 μM for 30 min), as indicated in the figure, and then processed and analyzed for eNOS membrane-positive immunostaining as described in the text. Each data point represents the mean ± SE derived from analysis of 100–120 cells per treatment in three independent culture preparations.
