Effect of brefeldin A on formation of OVA(257–264):K^b complexes by viable TAP1^−/− and TAP1^+/+ macrophages as assessed by T-cell assay. TAP1^−/− and TAP1^+/+ macrophages were cultured for 18 hr at 26° or 37° and then treated with or without brefeldin A (1 μg/ml) for 1 hr at the same temperature. The cells were then incubated with various concentrations of OVA(257–264) for 2 hr at the same temperature in the continued presence or absence of brefeldin A, washed, fixed, washed, and incubated with CD8OVA1.3 T-hybridoma cells, specific for OVA(257–264):K^b complexes.
