HP1 is required for normal levels of 3mK20-H4 in Drosophila. Whole cell extracts from wild type and Su(var)2–5 mutant larvae lacking HP1 were analyzed on immunoblots using H4-K20 methylation state-specific antisera. Extracts were also analyzed with antisera to HP1 to confirm the absence of HP1 in the mutant larvae and antisera to α-tubulin to ensure equivalent loading.
