In A-C, Kd(0) and δ were calculated from data from individual patches with 150 mm NaCl (A), sodium gluconate (B) or NaSCN (C) in the extracellular solution. D and E, the effect of changing the extracellular anion (from Cl− to gluconate (D) or from Cl− to SCN− (E)), quantified by the Kd(0) ratio as in Fig. 3D, is significantly reduced in all R334 mutants relative to wild-type (*P < 0.05; **P < 0.001). Mean of data from 3–5 patches in each case.
