In A-C, Kd(0) and δ were calculated from data from individual patches with 150 mm NaCl (A), sodium gluconate (B) or NaSCN (C) in the extracellular solution. D and E, the effect of changing the extracellular anion (from Cl to gluconate (D) or from Cl to SCN (E)), quantified by the Kd(0) ratio as in Fig. 3D, is significantly reduced in all R334 mutants relative to wild-type (*P < 0.05; **P < 0.001). Mean of data from 3–5 patches in each case.
