Efficient recognition of epitopes by RMA-S.B7–1 elicited CTLs requires the absence of TAP and the presence of MHC class I molecules in the target cell. B6 mice were immunized in vivo and splenocytes were restimulated in vitro with RMA-S.B7–1 cells and tested for cytotoxicity against (A) RMA-S (○) and RMA-S.TAP-2 (•) cells; (B) Con A blasts from B6 (□), TAP1−/− (▪), β2m−/− (▵), and TAP1/β2m−/− mice (▴); and (C) T2 (▿) and T2Kb (▾) cells.
